The Demon's Regimen is a Myth in Nioh 3. In The Demon's Regimen, the player must chase down Hayatemaru before they can be trained by Hattori Hanzo. Myths in Nioh 3 are story quests that consist of granting a request from the shadows of the deceased that can be completed to earn rewards. Myths are not required to reach the ending, but completing them will reward the player with various items that can help boost their character's progression or reveal secret locations.

Full The Demon's Regimen Mission Walkthrough
Chasing Hayatemaru
After unlocking The Demon's Regimen, head to the Hamamatsu Post - Terrace shrine [See Nioh 3 Map], then head northwest to the objective marker to find Hattori Hanzo. Speak with him, and he will ask you to chase Hayatemaru, a Scampuss. Pet Hayatemaru to start the chase, who will lead you down to an area where you will have to defeat three of Hanzo's underlings.
Duel with Hattori Hanzo
Once they are defeated, keep following Hayatemaru on the southern side. Follow the path under the bridge, and Hayatemaru will lead you to another three underlings of Hanzo. Defeat the enemies, then Hayatemaru will lead you to where Hattori Hanzo is. Once the dialogue with Hanzo is finished, you will fight him in a duel. Hattori Hanzo is a tanky boss who wields a spear, well-skilled at blocking attacks. For more information on how to defeat Hattori Hanzo, you may visit this page.
After Hanzo is defeated, talk to him to complete The Demon's Regimen. You will then be rewarded with a Ninja's Locks for completing The Demon's Regimen. You will also unlock The Demon's Regimen (Sub Mission) in the Battle Scroll for completing the myth, which will lead you straight to the duel with Hattori Hanzo.
