Nobunaga's Blade is a Sub Mission in Nioh 3. In Nobunaga's Blade, the player has to find Nobunaga's Blade, which the head priest of the Honnoji Temple has in their possession. Sub Missions in Nioh 3 are non-essential quests that can be completed in different chapters of the game. Sub Missions are not required to reach the ending, but completing them will reward the player with various rewards that can help boost their character's progression.
Oda Nobunaga was a hero of the Warring States period who fell at Honnoji Temple. It appears the head priest hid Nobunaga's favored sword someplace.

Search for Nobunaga's Blade
After unlocking Nobunaga's Blade, interact with any shrine and select the sub-mission to start it. You will be transported into an instance of Sanjo. Go forward up the steps and unlock the nearby shrine. Continue up the steps and you will find a kappa nearby a corpse you can loot. Follow the Kappa as it starts fleeing through a door to the west. There will be another door ahead of you that leads into an area with loot guarded by two enemy soldiers. After taking the loot, go back out and face north, defeat more enemies you encounter and loot the corpse at the corner before turning left.

Drop down the ledge to find a Mikoshi Nyudo and three enemy soldiers. Defeat the enemies in the area, then face south and go inside the structure with the blue lanterns to find a corpse you can loot. After looting the corpse, go north and then turn left. You will find the Kappa again. Follow the Kappa as it continues to flee past another gate where you will encounter a Wheelmonk and a Thorned Gaki.

Past the gate, turn left and defeat the yokai in the area. Keep an eye out for the Aberrant Soldier as it can shoot projectiles at you from a distance. After the area is cleared of yokai, cut the rope on the well to summon another Kappa. Continue forward and enter through the broken wall segment of the house and defeat the yokai inside. Loot the corpse in the area, then cross the chasm using the wooden platform to the west and continue following the Kappa.

Turn left after exiting the structure to find a shrine on the right side of a gate. Unlock the shrine then go through the gate and continue to follow the Kappa as it leads you to the final area.

Defeat the Yokai
Once you reach the area, a wave of yokai will appear. The first wave you will encounter will be a Thorned Gaki and a One-eyed Imp. After defeating both, the second wave will appear immediately, consisting of an Enki and a Gaki.

Once you defeat the Enki and the Gaki, collect their drops and the next pair of enemies will appear. The third wave will include the Red Kappa and a Kyokotsu. For this third wave, the Red Kappa is considered a boss and you can focus on defeating it first.

Once you defeat the Red Kappa, even if you haven't defeated the Kyokotsu yet, you will complete Nobunaga's Blade. You will unlock the title "Blessed by the God of Fortune". You will also unlock the guardian spirit Tengen Kujaku. Interact with the glowing orb in the arena to end the sub-mission. You will then receive Ninja's Locks and Swan Spear as completion rewards.

FYI you can actually kill the Kappa at anytime before it reaches the last area and the mission will end as well. It doesn’t require you to constantly stagger it to prevent it from escaping though.
