Those Left Behind is a Main Mission in Nioh 3. Those Left Behind is the objective that follows The Sweet Sound of Strings. Takechiyo must head towards Honnoji toward the Crucible to purify it after learning that Takasugi's ally, Oryo, has already made their way there. Objectives are special quests that are required to be progressed for players to unlock Main Missions. Main Missions in Nioh 3 are quests that are required to progress through the main story, which rewards the player with access to additional areas and levels to explore. A Main Mission will also give players various rewards for completing them.
Takasugi's ally, Oryo, has already started toward the Crucible that appeared in Honnoji, Takasugi will soon follow; I should head to Honnoji as well so that I may purify the Crucible.

Make Your Way Toward Honnoji Temple
After completing The Sweet Sound of Strings and speaking with Takasugi Shinsaku to complete A New Era, Those Left Behind will automatically start. In this mission, you are to make your way toward Honnoji Temple, which is currently engulfed by the Crucible. You can explore the other regions you pass through as you make your way toward Honnoji Temple to defeat more enemies and gain more Amrita to level up. You can refer to the map below.
After you reach the entrance to the Crucible, interact with it to enter, and the related main mission, Accursed Temple, will start. You will then have to make your way through the Honnoji Temple Crucible and purify it. To view the full walkthrough of the mission, click here.
After completing the main mission Accursed Temple and speaking with Takasugi Shinsaku and Himiko, now known as Oryo, you will also complete Those Left Behind and the next objective, Final Hope, will begin.
