Deadly Mark is a Weapon Skill from the Kusarigama Skill Tree in Nioh 3. Weapon Skills provide abilities that influence the character's combat moves and controls, which can be learned by going to the Acquire Martial Arts/Ninjutsu Menu. You can access this menu wherever you are in the game, and you need to spend either a Samurai Point or a Ninja Point to unlock weapon skills from their respective Skill Tree nodes. You can obtain Samurai and Ninja points by obtaining Samurai's Locks or Ninja's Locks.
