Every run of Slay the Spire 2 has a total of three Acts, each featuring a different locale/biome, and depending on how players path through each Act, they should be able to reach the final boss after 45–50 encounters. Depending on the types of rooms you enter and the build you're using, a typical Slay the Spire 2 run can last for up to 40 minutes, though runs can last for as long as an hour, or even more.

How Many Acts & Biomes Are There in Slay the Spire 2?
As mentioned earlier, there are three Acts in each Slay the Spire 2 run, but what players may not know is that each Act can be set in one of two different biomes. As of writing, only one alternate biome is available, and the other two are yet to be implemented in the game. The biomes we have in the game so far are:
Act 1 - Overgrowth
- Possible Bosses: Vantom, The Kin, Ceremonial Beast
The Overgrowth is the Spire's underbelly that's filled with weird and hostile critters. As the biome's name suggests, it's the most verdant of all the biomes, with backgrounds that typically feature grassy landscapes. Enemies in the Overgrowth are fairly basic considering the biome's purpose as an Act 1 area, and the same deal applies with its bosses:
- Vantom is protected by a destructible layer of ink that reduces damage taken to 1. Its attacks grow stronger with each turn, resetting after its third turn. Weak to multi-hit attacks, such as Regent's Stardust.
- The Kin involves a trio of enemies: one priest and two minions. The priest applies debuffs, while the minions buff themselves with Strength.
- The Ceremonial Beast is a simple enemy at first, but once its health drops, it will start inflicting some seriously debilitating debuffs on players.
Act 1 - Underdocks
- Possbile Bosses: Soul Fysh, Waterfall Giant, Lagavulin Matriarch
The Underdocks is a sewer-like area populated by all sorts of aquatic monsters. Its overall danger level is more or less the same as the Overgrowth, though the elite enemies and bosses players encounter here tend to be more troublesome.
- The Soul Fysh adds Beckon cards to the player's Draw Pile. If you have one in your hand, you will take damage at the end of your turn.
- The Waterfall Giant slowly builds up the Steam Eruption status on itself throughout the fight. When it dies, Steam Eruption will set off, dealing damage to players based on how many stacks it has. You need to have enough HP or Block to survive the eruption, otherwise, the run will fail. If you're playing Slay the Spire 2 with friends, only one of you needs to survive the blast.
- The Lagavulin Matriarch has a tendency to turtle up with high amounts of Block while building up Strength and inflicting Strength and Dexterity debuffs on players.
Act 2 - The Hive
- Possible Bosses: The Insatiable, Kaiser Crab, Knowledge Demon
The Hive is a significantly more dangerous area than either of the two Act 1 biomes. It's full of insects, and the elites here have a fairly good chance of beating players who come ill-prepared. The bosses are a bit trickier too.
- The Insatiable will instantly kill players in a set amount of turns, though this can be delayed by playing the Frantic Escape cards that the boss adds to your Draw and Discard Piles.
- Kaiser Crab involves a gimmick where you need to face the claw with the highest damage value. Players will take more damage from claws that hit them from behind.
- Knowledge Demon is a straight-up DPS check. Not only does the boss heal itself after a set number of turns, but it inflicts curses that have some seriously punishing effects.
Act 3 - Glory
- Possible Bosses: Doormaker, The Queen, Test Subject
Glory is part of the Spire's upper stratas, and this is where the most dangerous of enemies reside. Even a normal combat encounter here can spell the end of a run, and the elites and bosses found here are some of the deadliest in the game.
- Doormaker has a limited window of opportunity where players can deal any real damage to it. Once the window expires, it will retreat, forcing players to deal with a Door enemy that will always respawn with full health.
- The Queen inflicts heavy debuffs on players, and she starts the fight with a minion who deals a ton of damage.
- The Test Subject is a three-phase boss fight, with each fight featuring different mechanics that players need to deal with.
