You can start the Mouse PI for Hire Behind the Nuts side job after receiving One Flew, the main job that takes place at the Curdsville Nuthouse. After obtaining that objective, head to Tammy's Bearings behind the Soup to Nuts shop in the P.I. Office area and speak with Tammy.

Gloria is about halfway through One Flew, just after receiving the Jar-Head gun. Here is how to reach the Gloria location:

Look for a letter on top of a stack of papers in the room with the Jar-Head gun. This letter depicts the musical notes C-A-G-E.
Follow the main path until you run into Millie again.
Cross the bridge and look left to find a piano.
Play C-A-G-E on the piano keys.
Cross the bridge that drops and go right to find Gloria.
Snap a glorious photo of Gloria.
