Over the past decade, CZI has built technologies to observe and program biology, created large-scale data resources, and developed tools that accelerate discovery — all with a commitment to open science. With advances in AI, we’re entering the next evolution of our science work — and giving it a name that reflects both the history of our organization and our vision for the future: Biohub Moving forward, Biohub will be Mark and Priscilla’s primary philanthropic effort, dedicated to building frontier AI for frontier biology. For nearly a decade, our Biohub model has brought together world-class scientists and engineers to develop breakthrough tools. Now, we’re expanding that model — combining cutting-edge AI tools with large-scale compute dedicated to biological research. This positions Biohub to become the first research lab uniting AI and biology to accelerate discovery — and share everything it produces openly with the world. Science moves faster when people, tools, and data come together, and that’s exactly what Biohub makes possible. When my mom was a teenager, my grandparents made the unimaginable decision to send her on a boat across the South China Sea, guided by their faith that a better life was waiting on the other side. It might sound hard to believe, but I think about that act of courage nearly every day — and how it inspires me to build a better future for others through science. To me, innovation in science means giving brilliant minds the tools and freedom to explore the unknown — and translating discovery into hope. I’m so proud to lead Chan Zuckerberg Initiative and thrilled to see our groundbreaking science recognized by The Wall Street Journal Magazine.

We’ve been proud to support David Fajgenbaum, MD, MBA, MSc since the early days of our Rare As One Network. After surviving Castleman disease, David helped build a patient–researcher community that uncovered a treatment keeping him in remission for 15 years. Today, through the Castleman Disease Collaborative Network and Every Cure, he’s using AI to uncover new uses for existing drugs — opening up possibilities for treatments for people with rare diseases.
I recently had the chance to sit down with David Fajgenbaum, MD, MBA, MSc — a physician, scientist, and patient who’s transforming how we discover new treatments. We’ve been longtime fans and early supporters of David through the Rare as One Network. After surviving Castleman disease, David co-founded the Castleman Disease Collaborative Network to unite patients and researchers, and later launched Every Cure to systematically uncover new uses for existing drugs using AI. His work is a powerful example of what’s possible when patients lead, communities come together, and technology accelerates discovery. Proud to continue partnering with David as he brings new hope to the rare disease community.

The story highlights immunologist John Tsang, and #CZBiohubNY Investigator + Steering Committee member, whose research is helping reveal how the immune system changes with age, stress, and disease. Benchmarks are critical for AI to realize its full potential for accelerating biomedical discoveries. Our new tools let developers see how well their models really work, both technically and biologically, and help scientists pick the right model for their research. With seamless integration into your local environment, they will save developers time on model evaluation so they can spend more time building models that solve real biological questions. Freely available on CZI’s virtual cells platform, these benchmarking tools include vetted, high-quality tasks, metrics, and datasets from CZI, collaborators like NVIDIA Healthcare and the Allen Institute, and others in the single-cell community. CZI is leading the effort to develop and maintain these tools as a living, community-driven resource that will evolve as the field evolves, incorporating new data, refining metrics, and adapting to emerging questions in biology. By rallying the community around shared benchmarks, we’re speeding up scientific discovery and reshaping the way we study biology and understand disease.
